A tumor antigen peptide-loaded plasmacytoid dendritic cell (pDC) is a pDC that has been exogenously loaded with peptides derived from tumor antigens. This approach is used in cancer immunotherapy, where the goal is to harness the unique properties of pDCs to present tumor-specific antigens and stimulate an effective anti-tumor immune response. pDCs act as sentinels of the innate immune system, detecting pathogen-derived nucleic acids via endosomal Toll-like receptors (TLR7 and TLR9) and secreting IFN-I and other pro-inflammatory cytokines. When loaded with tumor antigen peptides, pDCs can process these peptides for presentation on MHC molecules, activating tumor-specific CTLs or helper T cells.
Presents tumor-associated peptides on MHC I/II molecules to activate cytotoxic T lymphocytes (CTLs) and helper T cells, leading to targeted killing of tumor cells.
