Overview

Tumor antigen presentation by dendritic cells (DCs) refers to the process in which DCs, as professional antigen-presenting cells, capture tumor-associated antigens and present them to T cells. This is a critical step for initiating and maintaining anti-tumor immune responses. DCs are uniquely equipped among immune cells to stimulate naive T cells, particularly through their ability to cross-present exogenous antigens on MHC class I molecules, thereby activating cytotoxic CD8+ T lymphocytes. The presence of functional intratumoral dendritic cells correlates with improved patient survival across several cancers due to enhanced stimulation of cytotoxic and helper T-cell responses. Dysfunctional or immature dendritic cells within tumors contribute significantly to immune evasion by failing to properly activate anti-tumor effector mechanisms. Several immunotherapeutic strategies leverage this biology including DC vaccines, which involve ex vivo-generated autologous dendritic cells pulsed with whole tumor lysates that elicit potent anti-tumor responses upon re-infusion into patients.
