Overview

Tumor antigen presentation describes the multi-step cellular machinery by which tumor-associated or tumor-specific antigens are processed and displayed on major histocompatibility complex (MHC) molecules at the cell surface for recognition by T cells. A competent antigen presentation pathway is fundamental for immune activation and effective anti-tumor T cell responses, but many cancers evade immune surveillance by disrupting this pathway. This machinery includes proteasomal degradation of intracellular proteins, peptide transport (e.g. via TAP), and MHC class I molecule assembly and surface expression. Defective presentation—via mutations, epigenetic silencing, or downregulation of MHC or pathway components—enables tumors to escape immune destruction. Therapeutic interventions aim to restore or augment antigen presentation and thus immune recognition, an underpinning of cancer immunotherapies such as checkpoint blockade, adoptive T cell transfer, and combinations with chemotherapy, radiotherapy, or epigenetic drugs[1][2][4][5][6].
