Overview

Tumor antigen presentation via major histocompatibility complex (MHC) on dendritic cells (DCs) is a critical process in the initiation and regulation of anti-tumor immune responses. Dendritic cells are professional antigen-presenting cells that capture, process, and present tumor-associated antigens to T lymphocytes, thereby orchestrating adaptive immunity against cancer. The efficiency of this process is closely linked to patient outcomes. Therapeutic strategies include ex vivo loading/pulsing of autologous dendritic cells with tumor lysates followed by reinfusion as vaccines.

Mechanism of action

Dendritic cells capture tumor antigens, process them into peptides, and present them on MHC molecules to activate T cells. This presentation can occur via MHC I (direct and cross-presentation to CD8+ T cells) or MHC II (classical presentation to CD4+ T cells), leading to cytotoxic T lymphocyte (CTL) responses or T helper cell-mediated immunity against the tumor.
