Tumor antigen-presenting dendritic cell (DC) activation of cytotoxic T lymphocytes (CTLs) refers to the process by which dendritic cells, acting as professional antigen-presenting cells, capture and process tumor antigens and present them via major histocompatibility complex class I (MHC I) molecules to naïve CD8+ T cells. This interaction is essential for priming and activating CTLs, which then acquire the ability to recognize and kill tumor cells displaying the same antigenic peptides. DCs are uniquely efficient at stimulating immunologically naïve T cells compared to other APCs. Upon activation, CTLs proliferate locally and differentiate into effector cells capable of killing target tumor cells. This mechanism forms the basis for several cancer immunotherapies, such as dendritic cell vaccines.
Ex vivo loading/maturation of patient-derived dendritic cells with tumor antigens followed by reinfusion aims to boost anti-tumor CTL activity (“dendritic cell vaccines”).
