Tumor-associated angiogenic endothelium refers to the specialized population of endothelial cells that line the blood vessels within a tumor microenvironment. Unlike normal quiescent endothelium, these cells are rapidly proliferating, structurally disorganized, and highly dependent on pro-angiogenic signaling pathways such as the VEGF/VEGFR axis [Nature Reviews Cancer, 2002]. They play a critical role in tumor progression by providing the necessary oxygen and nutrients for tumor growth and serving as a conduit for metastasis [Journal of Clinical Oncology, 2005]. Therapeutic strategies targeting this endothelium include anti-angiogenic agents, which prevent the formation of new vessels, and vascular disrupting agents (VDAs), which selectively destroy existing tumor vasculature [Clinical Cancer Research, 2011]. Targeting the tumor endothelium is a cornerstone of modern oncology, as it offers a way to "starve" the tumor and is less prone to the rapid mutational resistance seen in tumor cells themselves [Science, 1997].
