AGX-509 is an engineered T-cell receptor (TCR) therapy developed by AgonOx LLC that targets a specific, though currently undisclosed, tumor-associated antigen (TAA) [24, 33]. This TAA is presented on the surface of cancer cells in the context of the major histocompatibility complex (MHC), allowing the engineered T cells to recognize and eliminate malignant cells [15, 24]. The therapy is designed to treat solid tumors, potentially including melanoma, by leveraging the high specificity of TCR-mediated recognition [24, 27]. AGX-509 is part of a broader pipeline of immunotherapies that include tumor-infiltrating lymphocyte (TIL) based approaches and co-stimulatory agonists like OX40 [1, 17]. While the exact identity of the target antigen remains proprietary, it is classified as a tumor-associated antigen that is preferentially expressed in cancerous tissues compared to normal cells [24, 34]. The development of AGX-509 is currently in the preclinical stage, focusing on optimizing the TCR's affinity and the overall efficacy of the T-cell product [24, 32].
