Overview

Tumor-associated antigen B7 homolog 6 (B7-H6) is a type I transmembrane glycoprotein and a member of the B7 family of immune modulatory proteins.[2][3][4] It serves as a ligand for the natural cytotoxicity receptor NKp30, found on natural killer (NK) cells, and is typically not present or is undetectable in healthy tissues but is frequently expressed on a wide variety of human tumor cells, including hematologic malignancies and solid tumors.[2][3][4][5][6] The interaction with NKp30 activates NK cell cytotoxicity and cytokine response against tumor cells, playing a crucial role in tumor immunosurveillance.[3][4] Soluble forms of B7-H6 have been observed in the serum of cancer patients, where they can suppress NK-cell function and are associated with more aggressive disease and worse prognosis.[6][5][1] B7-H6 is under investigation as a therapeutic target, particularly for antibody-based immunotherapies, due to its selective expression on tumor cells and its involvement in modulating innate and adaptive immune responses.[7][3][6]
