Overview

Tumor-associated antigen-expressing malignant cells refer to cancer cells that display higher levels of specific proteins or other molecules known as tumor-associated antigens (TAAs) on their surfaces. While these antigens are elevated in cancers, they can also be present at lower levels on some normal cells. Targeting these cells is the basis of several immunotherapeutic strategies, including antibodies, CAR-T cells, and vaccines. However, not all TAAs are tumor-specific, leading to potential safety and efficacy challenges. This term does not denote a specific gene or protein but rather any malignant cell with TAA expression, which can vary widely depending on tumor type and antigen

02

Mechanism of action

Immune-mediated destruction of TAA-expressing cells, such as cytotoxic T lymphocyte activation, antibody-dependent cellular cytotoxicity, and vaccine-stimulated immunity

03

Biological functions

Cell proliferationImmune evasionCell signaling (via presentation of antigens)Other (context-dependent)
04

Disease associations

Cancer (all types of cancer with TAA expression)
05

Safety considerations

Off-tumor toxicity (because many TAAs are also expressed at low levels on normal cells)Autoimmunity (risk of immune response against normal tissues expressing the same antigen)Tumor heterogeneity and immune escape mechanisms, such as antigen loss variants
06

Interacting drugs

Cancer immunotherapies (such as checkpoint inhibitors, TAA-targeted monoclonal antibodies, TAA-targeted CAR-T cells, and cancer vaccines)
07

Biomarkers

Tumor-associated antigen positivity (varies by cancer, e.g., PSA, CEA, MAGE, mesothelin, AFP)
