Overview

The Tumor-associated antigen-MHC class II complex (TAA-MHC II) is a molecular assembly essential for the recognition of malignant cells by the adaptive immune system. Dendritic cells, acting as professional antigen-presenting cells, internalize tumor-derived proteins and process them into peptide fragments that are subsequently loaded onto MHC class II molecules for surface display (Roche & Furuta, 2015). This complex is specifically recognized by the T-cell receptor (TCR) on CD4+ T cells, including Cytokine-Induced Killer (CIK) cells, which play a pivotal role in orchestrating anti-tumor immunity (Jakel et al., 2014). Upon recognition, CD4+ T cells can provide necessary help to CD8+ cytotoxic T cells or exert direct anti-tumor effects through the secretion of cytokines like interferon-gamma or direct lysis of MHC II-positive tumor cells (Borst et al., 2018). In therapeutic contexts, this interaction is exploited through the development of dendritic cell vaccines and adoptive cell transfers, such as CIK cell therapy, which aim to enhance the frequency and activity of T cells specific for these complexes (Schmeel et al., 2014). However, the effectiveness of targeting TAA-MHC II can be limited by tumor-mediated downregulation of MHC molecules or the expression of inhibitory checkpoints that dampen T-cell activation (Vigneron, 2015).

Mechanism of action

The T-cell receptor (TCR) on CD4+ T cells or CIK cells binds specifically to the peptide-MHC II complex, triggering a signaling cascade that leads to T-cell activation, cytokine production (e.g., IFN-gamma), and direct or indirect tumor cell destruction.
