Tumor-associated antigen (TAA) presentation refers to the process by which antigens derived from tumor cells are processed and displayed on the cell surface, primarily via major histocompatibility complex (MHC) molecules, enabling recognition by T cells. This mechanism is central to immune surveillance and cancer immunotherapy. It enables the immune system—especially cytotoxic and helper T-cells—to recognize and target cancerous or mutated self-cells. Effective presentation of TAAs can lead to anti-tumor immunity, while defects in this process contribute to immune evasion and resistance mechanisms in tumors. Many immunotherapies—including checkpoint inhibitors—rely on effective antigen presentation for efficacy. Some tumors evade immunity by downregulating components of the antigen processing/presentation machinery (e.g., loss of MHC expression). Research is ongoing into drugs that restore or enhance antigen presentation as a means of overcoming resistance.
