Overview

Tumor-associated antigens presented via major histocompatibility complex class I molecules play a central role in anti-tumor immunity. Their presence enables effective recognition and elimination of malignant cells by cytotoxic T lymphocytes. However, many tumors develop mechanisms—such as downregulation/loss-of-function mutations—that impair this pathway as a means to evade immune destruction. Restoring or enhancing this pathway remains an important focus both for improving patient prognosis and increasing responsiveness to modern immunotherapies such as checkpoint blockade agents.
