The tumor-associated antigen-presenting pathway refers to the cellular processes by which tumor antigens are processed and displayed on the surface of cells, primarily via major histocompatibility complex (MHC) molecules, enabling recognition by T cells. This pathway is central to anti-tumor immunity and is a critical determinant of cancer immunotherapy efficacy. The pathway includes antigen processing by the proteasome, transport into the endoplasmic reticulum by TAP, peptide trimming by ERAP1/2, loading onto MHC class I molecules facilitated by tapasin, and presentation of the peptide-MHC complex on the cell surface. Defects in any component can lead to immune evasion. Interferons upregulate the pathway, while tumor microenvironment factors can suppress it. The integrity of this pathway is crucial for the effectiveness of immunotherapies.
