Overview

Tumor-associated antigen-specific autoantibodies (TAAbs) are immunoglobulins produced by the host's immune system in response to tumor-associated antigens (TAAs) that are mutated, overexpressed, or aberrantly modified in cancer cells (Tan, 2001, J Clin Invest). These autoantibodies serve as biological "reporters," as the immune system can amplify the signal of a small number of antigens, allowing for detection often months or years before clinical or radiological evidence of a tumor (Zaenker et al., 2016, Cancer Epidemiol Biomarkers Prev). While TAAbs are not therapeutic targets in the sense of being molecules that drugs are designed to inhibit or activate, they are critical biomarkers for early cancer detection, prognosis, and monitoring treatment response (Wang et al., 2023, Front Immunol). They have been extensively studied in various malignancies, including lung, breast, colorectal, and prostate cancers. The clinical utility of TAAbs is typically maximized by using multiplexed panels of antibodies to improve diagnostic sensitivity and specificity (MacDonald et al., 2017, J Thorac Oncol). Their presence reflects the body's natural humoral immune response to the "altered self" proteins characteristic of oncogenesis.
