Tumor-associated antigen (TAA)-specific cytotoxicity refers to the targeted killing of tumor cells by immune effector cells—primarily cytotoxic T lymphocytes (CTLs)—that recognize antigens preferentially or overexpressed on tumor cells, known as TAAs. This process involves CTLs binding to TAA-derived peptides presented on MHC molecules of tumor cells, leading to tumor cell lysis through various mechanisms including perforin/granzyme release and cytokine secretion. Targeting TAAs allows selective elimination of malignant cells, but challenges remain, including potential on-target/off-tumor toxicity and immune tolerance.
MHC-restricted recognition of TAA-derived peptides by CTLs, leading to perforin/granzyme-mediated lysis and/or cytokine secretion (IFNγ, TNFα) and FasL/Fas interaction inducing apoptosis.
