Overview

BRZ-201 is a preclinical mRNA-based cancer vaccine developed by Breeze Bio, Inc. (formerly GenEdit) using its proprietary NanoGalaxy platform [1, 2]. The vaccine utilizes highly-branched nanoparticles (HNPs) to deliver mRNA encoding tumor-associated antigens (TAAs) or personalized neoantigens directly to antigen-presenting cells (APCs) [15, 18]. This approach is designed to stimulate a robust and specific T-cell mediated immune response against malignant cells, particularly in "cold" tumors like glioblastoma that are typically resistant to immunotherapy [4, 5]. By encoding multiple antigens, BRZ-201 aims to overcome tumor heterogeneity and prevent immune escape [4]. The NanoGalaxy platform's non-viral delivery system is engineered to be less immunogenic and more tissue-specific than traditional lipid nanoparticles, potentially reducing systemic side effects [17, 18]. Preclinical studies have demonstrated the efficacy of this HNP-mRNA technology in eliciting potent antitumor immunity and improving survival in animal models of aggressive brain tumors [4, 5].

Mechanism of action

BRZ-201 delivers mRNA encoding tumor-associated antigens (TAAs) to antigen-presenting cells using highly-branched nanoparticles. The mRNA is translated into TAA proteins, which are then presented on MHC molecules to activate antigen-specific CD4+ and CD8+ T-cells, leading to the targeted destruction of tumor cells expressing those antigens [4, 18].
