Tumor-associated antigens recognized by LambdaGen multifunctional CARs represent the diverse set of surface molecules targeted by the proprietary immunotherapy platform developed by LambdaGen Therapeutics. The platform primarily focuses on B-cell lymphoma, utilizing multifunctional chimeric antigen receptors (m-CARs) that target multiple antigens simultaneously, such as CD19, CD20, and CD22, to enhance specificity and prevent tumor immune escape [1, 21]. These CARs are integrated into immune cells, including natural killer (NK) cells and monocytes/macrophages, using the company's Lambda-Integrase Genome Insertion Tool (LIGIT), which allows for the stable insertion of large multi-gene cassettes at safe harbor sites [3, 21, 28]. By targeting multiple antigens, these multifunctional CARs address the challenge of tumor heterogeneity and antigen downregulation, which are common causes of relapse in single-target CAR therapies [1, 12]. The use of iPSC-derived allogeneic cells further enables the development of 'off-the-shelf' products that aim to be more affordable and safer than traditional autologous CAR-T cell therapies [1, 5, 22]. This approach is being expanded to include solid tumor targets, leveraging the unique properties of different immune cell lineages to overcome the immunosuppressive tumor microenvironment [28, 31].
The multifunctional chimeric antigen receptors (CARs) are engineered to bind specifically to one or more tumor-associated antigens on the surface of malignant cells. Upon binding, the CAR triggers intracellular signaling pathways (typically involving CD3ζ and co-stimulatory domains like 4-1BB or CD28) that activate the engineered immune cells (such as NK cells or macrophages) to release cytotoxic granules, secrete pro-inflammatory cytokines, and induce targeted lysis of the tumor cells [1, 21, 25].
