The tumor-associated glycoprotein 72 (gp72) is a monomeric plasma membrane glycoprotein expressed on various human tumors, including osteogenic sarcoma, colorectal cancer, and ovarian cancer. It serves as both a tumor marker and a lymphocyte activation antigen. The antigen is specifically recognized by the murine monoclonal antibody 791T/36, which can be conjugated to toxins or cytokines for targeted anti-tumor therapy. Expression is substantially upregulated in neoplastic cells compared to low levels in normal tissues, particularly activated lymphocytes. Internalization following antibody binding allows intracellular delivery of therapeutics, though its route and rate of endocytosis are slower than classic receptors like transferrin. The antigen's restricted normal tissue distribution and upregulation in tumors enables its use in diagnostic immunolocalization and as a therapeutic target for cancer immunotherapy.
