Target intelligence / Profile preview
Tumor-associated glycoprotein 72 (TAG-72) is a high-molecular-weight, mucin-like oncofetal antigen that is overexpressed in a wide range of human adenocarcinomas, including colorectal, gastric, ovarian, breast, and pancreatic cancers [1, 6]. It is characterized by extensive O-linked glycosylation, specifically the sialyl-Tn (STn) epitope, which is recognized with high affinity by the second-generation monoclonal antibody CC49 [8, 14]. In normal adult tissues, TAG-72 expression is extremely limited, primarily found in secretory endometrium and rare gastrointestinal cells, making it a highly specific target for cancer therapy and diagnostics [4, 5, 13]. Therapeutically, TAG-72 is targeted using various modalities, including radioimmunotherapy, antibody-drug conjugates (ADCs), and chimeric antigen receptor (CAR) T-cell therapy [3, 5, 19]. The CC49 antibody and its humanized derivatives, such as minretumomab, have been extensively studied for their ability to deliver cytotoxic payloads or radioisotopes directly to tumor sites [2, 11]. Additionally, TAG-72 serves as a valuable biomarker; its serum form, CA 72-4, is used for monitoring disease progression and recurrence, while its tissue expression can guide intraoperative tumor resection through radioimmunoguided surgery [7, 15, 16]. Despite its promise, challenges such as tumor antigen heterogeneity and the development of human anti-mouse antibodies (HAMA) in response to murine-derived agents remain significant considerations in clinical development [10, 14].
Targeted delivery of cytotoxic agents, radioisotopes, or immune effector cells to TAG-72-expressing tumor cells, facilitating antibody-dependent cellular cytotoxicity (ADCC), radioimmunotherapy-induced DNA damage, or direct CAR-T cell-mediated lysis.
