Tumor cell lysis via activating receptor engagement refers to the process by which natural killer (NK) cells recognize and destroy tumor cells through the activation of specific surface receptors, primarily NKp30, NKp44, and NKp46 (Natural Cytotoxicity Receptors or NCRs). These receptors, upon engaging their ligands on tumor cells, initiate intracellular signaling cascades, leading to the formation of a cytolytic immune synapse, polarization of lytic granules, exocytosis of perforin/granzymes, and ultimately, tumor cell apoptosis. This pathway is a key target for cancer immunotherapy, particularly through the development of bispecific or multispecific antibodies that bridge NK cells and tumor cells for targeted killing.
Engagement of activating receptors (NKp30, NKp44, NKp46) on NK cells with ligands on tumor cells leads to immune synapse formation, polarization of lytic granules, exocytosis of perforin/granzymes, and induction of apoptosis in target tumor cells.
