Overview

NKG2D ligands (NKG2DLs) are a family of stress-induced cell surface molecules that are recognized by the activating immune receptor NKG2D. They are typically upregulated on tumor cells and trigger NK cell and T cell-mediated cytotoxicity. The main families include MICA/B and ULBP1-6. Tumor cells can evade immune detection by shedding soluble forms of NKG2DLs, which can impair antitumor immunity. Targeting the NKG2D/NKG2DL axis is an active area in cancer immunotherapy.
