The tumor cell replication machinery is a comprehensive term for the collective group of enzymes and proteins that execute DNA synthesis and cell division in malignant cells. This system includes the replisome—a complex of DNA polymerases, helicases, primases, and the sliding clamp (PCNA)—as well as topoisomerases and cell cycle regulatory kinases (Berdis, 2017). In cancer, this machinery is frequently overactive or dysregulated, leading to rapid proliferation and the accumulation of replication stress, which serves as a therapeutic vulnerability (NIH, 2024). Traditional chemotherapeutic agents, such as antimetabolites and alkylating agents, directly interfere with this machinery to induce DNA damage and apoptosis (PubMed, 2025). Additionally, oncolytic viruses and certain gene therapies like DeltaRex-G specifically hijack the high activity of this machinery in tumor cells to replicate their own genomes or express therapeutic transgenes (IntechOpen, 2024). While targeting these processes is effective in reducing tumor burden, it often results in significant side effects due to the shared reliance of normal proliferating cells on similar replication components (R Discovery, 2025).
