Tumor differentiation refers to the degree to which neoplastic cells resemble their normal counterparts in morphology and function (National Cancer Institute, 2024). In clinical practice, this status is used to 'grade' tumors, where well-differentiated tumors tend to grow slower than poorly differentiated or undifferentiated ones, which often demonstrate aggressive behavior (StatPearls, 2023). While not a specific molecule, 'differentiation therapy' targets biological pathways that inhibit maturation, such as the PML-RARα fusion protein in acute promyelocytic leukemia (APL) (Frontiers in Pharmacology, 2022). Drugs like tretinoin and enasidenib act by forcing these malignant cells to undergo terminal differentiation and lose their proliferative capacity (Molecules, 2022). Consequently, tumor differentiation represents a complex phenotypic endpoint and a pathological assessment rather than a single druggable receptor or enzyme (PMC3359654, 2012). This concept is fundamental to understanding tumor progression and the development of non-cytotoxic cancer treatments.
