Tumor-localized Tissue via Avidin–Biotin Interaction

Molecular classification
Protein-Small Molecule Interaction, Drug Delivery System
01

Overview

The avidin–biotin interaction is a high-affinity, non-covalent binding system used to target tumor tissues. A biotinylated targeting molecule (e.g., antibody) is administered, followed by a clearing agent (avidin/streptavidin) and then a biotinylated drug or imaging agent. The high affinity and multivalency of the interaction allow for enhanced tumor localization and payload delivery. It is not a therapeutic target in itself, but instead is a drug delivery system.

Mechanism of action

Pretargeting: Biotinylated targeting agent localizes to tumor, followed by avidin/streptavidin clearance, and subsequent administration of biotinylated payload.
