Tumor vascular disruption and necrosis refer to the targeted destruction of blood vessels within tumors, leading to a cascade of events that deprive tumor cells of oxygen and nutrients, ultimately resulting in cell death (necrosis). This process is both a natural consequence of rapid tumor growth outpacing its blood supply and an induced effect by certain therapeutic agents known as vascular disrupting agents (VDAs). VDAs selectively target proliferating endothelial cells in immature tumor vessels while sparing normal mature vasculature. Tumor necrosis is associated with aggressive disease features and metastatic potential.
