Overview

Tumor necrosis factor α-like ligand 1A (TL1A) is a member of the tumor necrosis factor superfamily (TNFSF15). It functions as a costimulatory signal for activated lymphocytes, particularly gut-specific CD4+ T cells, influencing Th1, Th2, and Th17 responses. TL1A interacts with its signaling receptor, Death Receptor 3 (DR3), to promote inflammation and fibrosis in autoimmune diseases like inflammatory bowel disease (IBD). Decoy Receptor 3 (DcR3) can bind TL1A and inhibit DR3 signaling. TL1A is considered a therapeutic target for chronic inflammatory conditions.
