Overview

Tumor necrosis factor alpha (TNF-alpha) is a pleiotropic pro-inflammatory cytokine belonging to the tumor necrosis factor superfamily (UniProt P01375). It is primarily secreted by macrophages but also by T-cells, natural killer cells, and mast cells in response to stimuli such as bacterial lipopolysaccharides (StatPearls: TNF Inhibitors). TNF-alpha acts as a master regulator of the inflammatory response by binding to its receptors, TNFR1 and TNFR2, which triggers various intracellular signaling pathways including the NF-kappaB and MAPK cascades (PubMed: PMC3108497). These pathways promote the expression of other pro-inflammatory cytokines, adhesion molecules, and enzymes involved in tissue destruction (StatPearls). Overproduction of TNF-alpha is a hallmark of several chronic inflammatory and autoimmune disorders, making it a high-value therapeutic target. Monoclonal antibodies and soluble receptor fusion proteins that neutralize TNF-alpha have revolutionized the management of diseases like rheumatoid arthritis and Crohn's disease by effectively suppressing the systemic inflammatory cascade (NIH: MedlinePlus).
