Tumor necrosis factor alpha-induced protein 8-like protein 1 (TIPE1/TNFAIP8L1) is a cytoplasmic protein family member expressed in various tissues except mature B and T lymphocytes[1]. It functions mainly as a regulator of cell apoptosis, acting as a tumor suppressor by inhibiting cell proliferation, EMT, and metastasis, and promoting apoptotic pathways in most cancers. Structurally, TIPE1 features a conserved domain with several alpha helices forming a hydrophobic cavity[1][2][4]. It negatively regulates mTOR signaling, contributing to its roles in inflammation and metabolic regulation[3][6][7]. Aberrant expression or dysfunction of TIPE1 is implicated in cancer progression, tissue inflammation, and fibrosis, marking it as a potentially valuable biomarker and therapeutic target, particularly in oncology[1][5][7].
TIPE1 exerts its mechanism of action through mTOR inhibition, which may be a target for drugs acting via mTOR pathway blockade. It also promotes apoptosis, representing a potential mechanism for anticancer therapeutics leveraging its activity. Furthermore, it inhibits cell proliferation and metastasis through the downregulation of EMT, JNK, p65, ERK phosphorylation, and MMPs.
