Overview

The lectin-like domain of TNF-α is a specific region within the TNF-α protein (residues Cys101-Glu116 in humans) that binds to certain oligosaccharides. This domain is spatially distinct from the receptor-binding sites of TNF-α and exerts unique biological effects, including the activation of epithelial sodium channels (ENaC) and modulation of inflammation. Synthetic peptides mimicking this domain, such as solnatide/TIP peptide, are being developed as therapeutics for acute lung injury and other inflammatory conditions. The mechanism involves binding to glycosylated membrane components of ENaC, followed by uptake into lipid rafts and direct activation of the channel, leading to increased sodium and fluid clearance.
