Overview

The TNF-α production pathway encompasses the synthesis, processing, and secretion of TNF-α, a key cytokine in immune regulation. TNF-α is produced by several cell types—primarily macrophages, but also T cells, natural killer cells, dendritic cells, fibroblasts, and others. Its gene is located on chromosome 6 and regulated by transcription factors like NF-κB. TNF-α is initially synthesized as a transmembrane precursor (tmTNF-α), which is cleaved by the TNF-α-converting enzyme (TACE) to release the soluble form (sTNF-α). Both forms act by binding to two receptors—TNFR1, expressed on most tissues and mediating cytotoxic and apoptotic pathways, and TNFR2, more selectively expressed on immune cells, mediating survival and inflammatory responses. Downstream signaling leads to activation of transcription factors (NF-κB, AP-1), caspase cascades, and production of other cytokines, culminating in inflammation, cell death, or proliferation. Dysregulation of this pathway plays a central role in the pathogenesis of many diseases, particularly chronic inflammatory, autoimmune, and malignant conditions.
