The Tumor Necrosis Factor (TNF) ligand superfamily (TNFSF) comprises a group of approximately 19 ligands that are essential regulators of immune homeostasis, inflammation, and development [UniProt: Family: TNF_ligand_superfamily]. These proteins are primarily type II transmembrane proteins that form bioactive homotrimers, which interact with corresponding receptors in the TNF receptor superfamily (TNFRSF) to trigger diverse cellular responses including survival, differentiation, and apoptosis [PMID: 12776174]. Key members such as TNF-alpha (TNFSF2), RANKL (TNFSF11), and BAFF (TNFSF13B) are central to the pathophysiology of autoimmune disorders like rheumatoid arthritis, bone loss in osteoporosis, and B-cell malignancies [PMID: 15561100]. Pharmacological intervention typically involves the use of monoclonal antibodies or soluble decoy receptors to neutralize these ligands, effectively dampening overactive immune responses [DrugBank: DB00003, DB00051]. Despite their therapeutic success, targeting these molecules poses significant safety challenges, most notably the risk of opportunistic infections due to the suppression of critical host defense mechanisms [PubMed: 21248325].
Neutralization of specific TNFSF ligands to prevent binding to their cognate TNFRSF receptors, thereby inhibiting downstream pro-inflammatory or pro-apoptotic signaling pathways such as NF-kappaB and MAPK [PMID: 12776174, PMID: 15561100].
