TNFSF12, also known as TWEAK, is a cytokine belonging to the tumor necrosis factor (TNF) ligand superfamily. It exists in both membrane-bound and secreted forms. It primarily binds to the FN14/TWEAKR receptor. TWEAK has several key biological activities including induction of apoptosis, promoting angiogenesis, and mediating activation of the NF-kappa-B signaling pathway. Excessive activation of the TWEAK/FN14 pathway has been implicated in pathological tissue changes during chronic injury, such as chronic inflammation and fibrosis.
