Overview

Tumor necrosis factor messenger RNA (TNF mRNA) is the genetic transcript that encodes the Tumor Necrosis Factor-alpha (TNF-alpha) protein, a key pro-inflammatory cytokine and a member of the TNF superfamily. TNF-alpha is primarily produced by activated macrophages and plays a critical role in orchestrating the innate immune response, regulating cell death, and inducing the expression of other inflammatory mediators (UniProt P01375). Dysregulation and overproduction of TNF-alpha, driven by high levels of its mRNA, are central to the pathogenesis of numerous autoimmune and inflammatory conditions, such as rheumatoid arthritis and inflammatory bowel disease (PMID: 16169925). While traditional biologics target the secreted protein, therapeutic strategies targeting the mRNA transcript—such as antisense oligonucleotides (e.g., ISIS 104838) and siRNAs (e.g., STNM01)—aim to silence the gene's expression at the source (PMID: 12490586, PMID: 25352185). By promoting the degradation of TNF mRNA or inhibiting its translation, these therapies reduce the overall synthesis of the cytokine, offering a potent approach to managing chronic inflammatory states.
