Overview

Tumor necrosis factor receptor 1 (TNFR1) signaling complex II is a cytoplasmic multi-protein assembly that mediates programmed cell death in response to TNF-alpha stimulation. It typically forms after the internalization of the TNFR1 receptor and the subsequent dissociation of the membrane-bound, survival-promoting Complex I (Micheau & Tschopp, 2003). Complex II is composed of core proteins including Fas-associated death domain (FADD), pro-caspase-8, and Receptor-interacting serine/threonine-protein kinase 1 (RIPK1). Depending on the cellular context and the presence of inhibitors like c-IAPs or c-FLIP, this complex can trigger either apoptosis via caspase-8 activation or necroptosis via the recruitment of RIPK3 and MLKL (Vandenabeele et al., 2010). Because of its central role in determining cell fate, Complex II is a major focus for drug development; SMAC mimetics are designed to promote its formation to kill cancer cells, while RIPK1 inhibitors aim to block its activity to treat chronic inflammatory and neurodegenerative conditions (Annibaldi & Silke, 2018; Yuan et al., 2019).
