Overview

Tumor necrosis factor receptor 2 (TNFR2) is a membrane-bound receptor protein and a member of the tumor necrosis factor receptor superfamily. It contains four cysteine-rich domains in its extracellular region for ligand binding, primarily membrane-bound tumor necrosis factor-alpha (tmTNF). Unlike TNFR1, TNFR2 lacks an intracellular death domain and primarily signals for cell survival, proliferation, immune modulation, or tissue regeneration through activation of both canonical and non-canonical NF-κB pathways. TNFR2 is expressed on endothelial cells, fibroblasts, subsets of neurons, and immune cells, notably regulatory T cells and myeloid-derived suppressor cells. It is implicated in tumor progression, autoimmune diseases, and other conditions due to its dual pro-inflammatory/anti-inflammatory roles. Therapeutic strategies are being developed to selectively target the stimulatory or inhibitory functions of this receptor for cancer immunotherapy or treatment of autoimmune diseases.
