Tumor necrosis factor receptor-associated factors (TRAFs) are a family of structurally related intracellular adaptor proteins that play central roles in signal transduction from members of the tumor necrosis factor receptor (TNFR) superfamily and various other immune receptors. They regulate inflammation, antiviral responses, apoptosis, and cell survival. Dysregulation or mutation within this family is implicated in autoimmune/inflammatory diseases, immunodeficiency, and cancer progression.
TRAFs function as adaptor proteins by associating with the cytoplasmic tails of activated receptors, mediating downstream signaling events like the activation of NF-κB and MAPK pathways.
