Overview

Tumor necrosis factor receptor-associated factor 6 (TRAF6) is a cytoplasmic adapter protein and signal transducer that plays a central role in multiple signaling pathways, particularly those involved in immune and inflammatory responses. It functions as an E3 ubiquitin ligase, catalyzing the formation of K63-linked polyubiquitin chains, which serve as scaffolds for recruiting other signaling molecules. TRAF6 is essential for activating downstream signaling cascades, including the NF-kappaB and JNK/MAPK pathways, regulating inflammation, immune responses, cell survival, differentiation, and apoptosis. Mutations or dysregulation in TRAF6 have been linked to several diseases, including Ectodermal Dysplasia A10, Osteopetrosis, immunological disorders, cancer, and inflammation.
