Overview

Tumor necrosis factor receptors (TNFRs) are a family of cell surface receptors that bind tumor necrosis factor (TNF), a key cytokine involved in inflammation, immune regulation, and cell death. The two main TNF receptors are TNFR1 and TNFR2. They mediate both beneficial effects (anti-infection/tumor immunity) and detrimental ones such as systemic inflammatory response syndrome when dysregulated. In cancer biology they can promote either anti-tumor immunity or support tumor progression depending on context. Excessive activation/signaling through these receptors underlies many chronic inflammatory diseases—hence anti-TNF therapies target this axis effectively.
