Overview

Tumor necrosis factor receptor superfamily member 18 (TNFRSF18), also known as glucocorticoid-induced TNFR-related protein (GITR or CD357), is a type I transmembrane receptor in the TNF receptor superfamily predominantly expressed on activated T cells and regulatory T cells (Tregs)[1][2][3][4]. Engagement of GITR modulates immune responses by promoting T-cell activation, proliferation, and cytokine production, while abrogating Treg suppressive function. GITR signals via recruiting TRAF family adaptor proteins, leading to activation of NF-κB and MAPK pathways. GITR plays context-dependent roles in control of immune tolerance and inflammation, and its activation or inhibition represents a promising strategy in cancer immunotherapy and the treatment of autoimmune diseases. Its biological importance and therapeutic relevance stem from its capacity to fine-tune immune activation and tolerance[1][2][3][4].
