Tumor necrosis factor receptor superfamily member 19L (RELT) is a type I transmembrane orphan receptor within the tumor necrosis factor receptor superfamily. RELT is most highly expressed in hematopoietic tissues, particularly lymphoid tissues, and has two paralogs (RELL1 and RELL2). Structurally, RELT has an extracellular domain (ECD) with cysteine-rich regions but does not bind any known TNF family ligands and lacks the classic death domain typical of some TNFRSF members. Functional studies show RELT can activate the NF-κB signaling pathway and induce apoptosis, though via mechanisms distinct from classic death receptors like TNFR1 and Fas. RELT and its family members (RELL1, RELL2) play roles in immune modulation, inflammation, and cell death, and have been implicated in cancer pathobiology (sometimes risk, sometimes protective), heart disease, and other conditions. The endogenous ligand and precise physiological functions of RELT require further elucidation, but emerging research underscores its importance in diverse biological and pathological processes[1].
