Overview

Tumor Necrosis Factor Receptor Superfamily Member 25 (TNFRSF25), also known as DR3, is a type I transmembrane glycoprotein and a member of the TNF receptor superfamily. It is primarily expressed on activated T lymphocytes, FoxP3+ regulatory T cells, and other immune cells. Its only known ligand is TL1A, which is produced by antigen-presenting cells. Upon binding TL1A, DR3 can activate NF-kappaB signaling, leading to increased IL-2 responsiveness and proliferation of T cells, or it can trigger apoptosis through the FADD/caspase cascade. DR3 plays a role in immune regulation, lymphocyte homeostasis, and is implicated in the pathogenesis of autoimmune diseases and inflammatory bowel disease.
