Overview

Tumor necrosis factor receptor superfamily member 9 ligand (4-1BBL), also known as CD137L or TNFSF9, is a type II transmembrane glycoprotein belonging to the TNF ligand superfamily. It is expressed primarily on antigen-presenting cells (APCs) and some carcinoma cell lines. 4-1BBL binds to its receptor 4-1BB (CD137) on T lymphocytes, acting as a costimulatory molecule to enhance T cell activation, survival, and proliferation. This interaction plays a crucial role in regulating immune responses, particularly in cytotoxic CD8+ T cell activity. Agonists targeting the 4-1BBL/4-1BB pathway are being investigated for cancer immunotherapy due to their ability to boost anti-tumor immunity.
