Overview

Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand (TRAIL) is a cytokine that induces selective tumor cell death through activation of the extrinsic apoptotic cascade via binding to pro-death receptors DR4/TRAIL-R1 & DR5/TRAIL-R2. Decoy receptors DcR1/TRAIL-R3 & DcR2/TRAIL-R4 act as negative regulators. Signaling is primarily pro-apoptotic but can trigger non-canonical survival/motility pathways depending on context. It is an attractive anticancer target due to selectivity; however, clinical translation has been challenged by resistance mechanisms.
