Tumor Necrosis Factor-like Cytokine 1A (TL1A), also known as TNFSF15, is a type II transmembrane protein belonging to the TNF superfamily. It exists as both membrane-bound and soluble forms and plays a critical role in immune regulation, influencing the proliferation, activation, differentiation, and apoptosis of immune cells. TL1A signals primarily through its receptor DR3/TNFRSF25, activating MAPK, NF-kB, and caspase-dependent pathways. It is implicated in various autoimmune and inflammatory diseases, including IBD, rheumatoid arthritis, and asthma, making it a therapeutic target. Anti-TL1A antibodies are under clinical investigation as potential treatments for these conditions.
