Overview

NHS76 is a fully human monoclonal IgG1 antibody developed for targeting solid tumors by binding to intracellular antigens—specifically, single and double-stranded DNA that becomes accessible in areas of tumor cell death (necrosis) within solid tumors. Unlike most antibodies that bind extracellular targets, NHS76 recognizes nuclear material only present outside cells when membrane integrity is lost due to necrosis or apoptosis. This unique property allows it to selectively deliver therapeutic agents—such as interleukin‑12 in the fusion protein NHS–IL12—directly into the immunologically active environment within tumors while sparing healthy tissue. The approach has been shown preclinically and clinically to enhance anti-tumor immune responses by increasing T-cell infiltration and activity within tumors, especially when combined with other immunotherapies or vaccines[2][3][4][5].
