Tumor Protein p53 (Mutant) (mutant p53)

Target
mutant p53
Molecular classification
Transcription factor, Tumor suppressor
01

Overview

Mutant p53 refers to tumor protein p53 with mutations, most commonly missense mutations in the DNA-binding domain. These mutations lead to loss of normal tumor suppressor functions, such as cell cycle arrest and apoptosis, and often confer new oncogenic gain-of-function properties, including promoting tumor progression, metastasis, angiogenesis, and metabolic reprogramming. Mutant p53 is a major therapeutic target in cancer.

Mechanism of action

Varies depending on the specific mutant p53 and therapeutic approach; strategies include restoring wild-type p53 function, inhibiting mutant p53 gain-of-function activities, or targeting downstream pathways affected by mutant p53.
