Tumor protein p53 binding protein 2 (TP53BP2), also known as apoptosis-stimulating of p53 protein 2 (ASPP2), is a multifunctional tumor suppressor and apoptosis regulator encoded by the TP53BP2 gene. It belongs to the ASPP family and is characterized by containing four ankyrin repeats and an SH3 domain, facilitating multiple protein-protein interactions. TP53BP2 directly binds the tumor suppressor p53 and its family members, enhancing their ability to induce apoptosis by upregulating pro-apoptotic gene transcription. It localizes to the perinuclear region and modulates cell fate decisions notably in response to cellular stress. Its biological relevance is highlighted in a range of human cancers, where loss or inactivation may contribute to tumor development and progression.
