The myeloma idiotype protein is the unique, patient-specific variable region of the monoclonal immunoglobulin (M-protein) produced by malignant plasma cells in multiple myeloma (Source: PubMed, PMID: 10472885). Each clone of malignant B-cells expresses a distinct idiotype, which functions as a tumor-specific antigen because it is not found on normal B-cells or other tissues (Source: NCI Thesaurus). In therapeutic applications, this protein is utilized as the primary component of idiotype vaccines, where it is often conjugated to an adjuvant like Keyhole Limpet Hemocyanin (KLH) to enhance its immunogenicity (Source: Blood, 1992, 80(9):2215-2220). The goal of such therapy is to stimulate the patient's own T-cells and B-cells to recognize and eliminate the cancerous plasma cell population (Source: Journal of Clinical Oncology, 2011, 29(20):2787-2794). Beyond its role as a target, the protein serves as a critical biomarker; its concentration in the blood or urine is used to diagnose the disease and monitor the effectiveness of chemotherapy or stem cell transplantation (Source: International Myeloma Working Group).
