Overview

The tumor-specific immunoglobulin idiotype (Id) refers to the unique antigenic determinants found in the variable regions of immunoglobulins (antibodies) produced by malignant B cells, such as those in B-cell lymphomas and multiple myeloma. Each B-cell tumor expresses a clonal immunoglobulin with a distinct idiotype, which is not present on normal cells. These idiotypes are primarily determined by the complementarity-determining regions (CDRs) of the variable domains of both heavy and light chains, often shaped by somatic mutations. It can be recognized by both antibodies and T-cells, making it an effective target for immune-based therapies. Tumor-specific idiotypes have been exploited as targets for active immunotherapy, particularly in vaccine development.
