Tumor-specific mutant peptide antigens (“neoantigens”) are uniquely mutated short peptides generated by genomic alterations exclusive to tumors. They serve as potent targets for personalized immunotherapies due to their high specificity and ability to elicit robust anti-tumoral immune responses without affecting healthy tissue.
Neoantigens are presented on MHC molecules and recognized by T cells, leading to targeted killing of tumor cells. Personalized vaccines prime the immune system against these neoantigens. Adoptive cell therapies utilize T cells engineered to target specific neoepitopes. Immune checkpoint blockade efficacy is enhanced in tumors with high neoantigen load.
